番茄红素对高脂性脂肪肝大鼠的保护作用及其机制研究

中文摘要:目的 研究番茄红素(lycopene,LP)对高脂性脂肪肝大鼠的保护作用,并探讨其可能的作用机制。方法 取120只实验用大鼠随机分为正常对照组、模型组、LP(10、20、40mg/kg)治疗组和辛伐他汀1.8mg/kg治疗组(阳性对照组)。通过高脂饲料喂养8周制备高脂性脂肪肝大鼠模型;灌胃给药治疗,每天1次,疗程4周。治疗完成后,测定大鼠体质量、肝重量并计算肝脏指数;测定肝功能指标(ALT、AST、ALP、CHE);HE染色观察肝脏组织结构形态学变化;检测血脂(TC、TG、LDL-C、HDL-C)含量,游离脂肪酸(FFA)含量,肝脏组织中抗氧化酶(SOD、GSH-Px、T-AOC)活性和丙二醛(MDA)含量,血清中炎性因子(TNF-α、IL-6)水平。结果 与模型组比较,LP(20、40mg/kg)治疗组大鼠肝重量和血清中ALT、AST、ALP、TC、TG、FFA、TNF-α、IL-6含量均显著降低,肝脏组织中SOD活性显著升高且MDA含量显著降低;LP(40mg/kg)治疗组肝脏指数均显著降低,血清中CHE、LDL-C含量显著降低且HDL-C含量显著升高,肝组织中GSH-Px和T-AOC活性均显著升高,上述差异均有统计学意义(P<0.05);LP各治疗组大鼠肝脏组织病变呈不同程度改善,以LP40mg/kg治疗组效果最为显著。结论 LP对高脂性脂肪肝大鼠具有保护作用,其机制可能与LP能够有效降低血脂、改善抗氧化酶活性、抑制氧化应激损伤、降低炎性介质水平有关。

Protective Effects and Mechanism of Lycopene on Hyperlipidemic Fatty Liver in Rats
Abstract:Objective To investigate the protective effects and mechanism of Lycopene(LP) on hyperlipidemic fatty liver in rats. Methods One hundred and twenty experimental rats were randomly devided into six groups: normal control group, model group, LP(10, 20, 40mg/kg)ttreated groups and Simvastatin 1.8mg/kg treated group(positive control group). The rat models were made by feeding with high fat diet for eight weeks. The drugs were given by intragastric administration for four weeks. And then, the body weight and hepatic index were detected. The content of ALT, AST, ALP, CHE, TC, TG, LDL-C, HDL-C, FFA, TNF-α and IL-6 in serum were determined. The histopathological changes of hepatic tissue was observed by HE staining. The activity of SOD, GSH-Px, T-AOC and the content of MDA in hepatic tissue were determined. Results Compared with the model group, the body weight of LP(20, 40mg/kg) treated groups were significantly decreased (P<0.05), the content of ALT, AST, ALP, TC, TG, FFA, TNF-α, IL-6 in serum were significantly decreased (P<0.05), the activity of SOD in hepatic tissue was significantly increased and the content of MDA were significantly decreased(P<0.05). The hepatic index in LP 40mg/kgttreated group was significantly decreased, the content of CHE, LDL-C in serum were significantly decreased and the content of HDL-C was significantly increased, the activity of GSH-Px, T-AOC were significantly increased, and all of the difference above were significant(P<0.05, P<0.01). The histopathological changes of hepatic tissue in LP treated groups were significantly improved, especially in the LP 40mg/kgtreated group. Conclusion LP had protective effects on hyperlipidemic fatty liver in rats, which perhaps related to its effects of reducing cholesterol, improving the activity of antioxidant enzymes, inhibiting oxidative stress, lowering the level of TNF-α and IL-6.
