中文摘要:目的 姜黄素类似物D33对同种异体肢体移植原位再植模型大鼠的免疫功能的作用及机制。方法 构建SD/Wistar大鼠的同种异体肢体移植原位再植模型,随机分为对照组和D33组,D33组每日腹腔注射20mg/kg D33,对照组注射等体积生理盐水。另选择正常Wistar大鼠作为正常组。流式细胞术分析各组T细胞亚型分布以及血液中炎性因子TNF-α、IL-1、IL-2、IFN-γ、IL-4、IL-5和IL-10 的含量。从D33组大鼠的胰岛和淋巴组织中分别分选受者来源DC细胞和供者来源DC细胞,Western blot法检测DC细胞中NF-κB、p38/MAPK、ERK/MAPK和Ⅰ型 IFN(IFN-α/β)通路的激活情况。结果 D33的大鼠的移植手术效果较佳。D33组受鼠血液中CD4+、CD8+、CD4+/CD8+的比例均低于对照组,但高于正常组。D33组中TNF-α、IFN-γ、IL-1、IL-2和IL-5水平均低于对照组,而IL-4和IL-10水平高于对照组。受者来源DC细胞中,D33可明显抑制NF-κB、p38/MAPK、ERK/MAPK和Ⅰ型IFN(IFN-α/β)通路的激活,在供者来源DC细胞中,D33的影响并不明显。结论 D33通过抑制TLR4相关通路诱导机体免疫耐受。 |

Effects and Mechanisms of Curcumin Anologues D33 on Immune Tolerance in Rats |

Abstract:Objective To study the effect and molecular mechanism of immune tolerance induced by curcumin analogues D33. Methods SD/Wistar rats homologous allograft limb transplantation in situ replantation model was constructed and randomly divided into D33 group and control group.Rats in D33 group was intraperitoneally injected with 20mg/kg D33, and ratsin control group was injected with equal volume of saline. The normal Wistar rats were selected as normal group. The distribution of T cells and the contents of inflammatory cytokines TNF-α, IL-1, IL-2, IFN-γ, IL-4, IL-5, IL-10 were analyzed in flow cytometry. Dendritic cell (DC) was separated from the D33 rats islets and lymphoid tissue. Western blot was used to detect the activation of NF-κB, p38 and ERK/MAPK and type Ⅰ IFN (IFN-α/β) pathway. Results The effect of the transplantation of rats with D33 was better. The proportion of CD4+, CD8+, CD4+/CD8+ in the blood of the D33 group rats was lower than the control group, but higher than the normal group. The levels of IL-5, IFN-γ, IL-1, IL-2 and TNF-α in the D33 group were lower than those of the control group, while the levels of IL-10 and IL-4 were higher than those of the control group. In DC cells of receptor rats, D33 significantly inhibited the activation of NF-κB, p38/MAPK, ERK/MAPK and IFN (Ⅰ) pathway, and the effect of D33 was not obvious in donor DC cells. Conclusion D33 induced immune tolerance by inhibiting TLR4 pathway activation. |
