法舒地尔对小鼠造影剂急性肾损伤的影响

中文摘要:目的 探讨法舒地尔是否可以通过抑制Rho/ROCK信号通路,减轻氧化应激损伤及炎性反应,从而减轻造影剂急性肾损伤的发生。方法 30只小鼠行单侧肾蒂结扎术,术后1周,随机分为模型组、法舒地尔干预组或对照组(n=10/组)。小鼠经尾静脉注射碘克沙醇注射液(4.0g I/kg)或等剂量的生理盐水(对照组)。法舒地尔干预组在注射碘克沙醇前12h、2h以及注射后4h 3个时间点给予法舒地尔(10mg/kg,腹腔注射)。造模后24h检测小鼠血清肾功能标志物、炎症、氧化应激相关蛋白的表达水平。结果 与模型组相比,法舒地尔可以显著的降低血清Cr水平,减少肾组织活性氧簇(ROS)的产生及其诱导的DNA损伤。Wester blot法检测结果显示,法舒地尔可以显著减少炎性细胞因子TNF-α、IL-6蛋白水平(P<0.05)。结论 法舒地尔可以改善造影剂诱导的血清肌酐升高,其肾脏保护作用机制主要是通过抑制Rho/ROCK信号通路,从而发挥抗炎、抗氧化应激作用。

Fasudil Attenuates Contrast-induced Acute Renal Failure
Abstract:Objective To examine the effects of fasudil, a Rho kinases inbibitor, on the treatment of the contrast-induced acute renal failure in the mouse model. Methods Thirty mice were subjected to unilateral ligation of the left anterior renal pedicle. Seven days later, animals were divided into three groups as follows:(1) control group, (2) contrast media group (CM group), (3) Fasudil group(FS group). Animals from group 2-3 received iodixanol (4.0g iodine/kg), the control group received saline. At 12h, 2h before injection of iodixanol and at 4h after this operation, animals in groups 3 received 10mg/kg fasudil. Renal function parameters, the expression of oxidative stress injury proteins,inflammation regulatory proteins, were determined 24 hours later. Results Fasudil inhibited inflammation reaction induced by CM treatment (P<0.05), and notably ameliorate contrast medium induced medullary damage, restore renal function, and suppress renal tubular oxidative stress injury. Conclusion The results showed that fasudil attenuated CM-induced rise of creatinine suggesting its nephroprotective effect, which was partly attributed to it's anti-inflammatory and anti-oxidant effects by inhibiting the Rho/ROCK pathway.
