中文摘要:目的 检测G0S2基因在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的甲基化率及表达量,并探讨其成为NSCLC早期诊断和预后预测特异性分子标志物的可能性。方法 采用焦磷酸测序法测定组织细胞中G0S2基因甲基化率,荧光定量PCR测定组织细胞中G0S2基因mRNA的表达量,根据NSCLC患者的人数平均分为两组后,cut-off值为0.2,以此分为G0S2基因高表达组与低表达组,并对G0S2基因表达量的影响因素进行单因素分析,并分析该基因表达量与患者预后之间的关系。结果 在NSCLC患者中G0S2基因存在高甲基化率(40%),并且该基因在正常组织中的表达量与肿瘤组织中的表达量比较差异有统计学意义(P=0.000);在单因素分析中发现NSCLC患者的年龄、性别、吸烟史、肿瘤的病例类型、分期分级对G0S2基因的表达量均无明显影响(P>0.05);G0S2基因的表达量与NSCLC患者的预后无明显相关性(P>0.05)。结论 检测G0S2基因的表达情况对NSCLC患者早期诊断有一定的作用,但在后续指导肺癌分期、预测患者预后等方面无明显参考价值。 |

Epigenetic Changes of G0S2 Gene in NSCLC Patients |

Abstract:Objective To evaluate the value of methylation levels and expression of G0S2 gene in early diagnosis and predicting prognosis of non-small cell lung cancer (NSCLC). Methods We used pyrosequencing to detect G0S2 methylation levels and reverse transcription PCR to detect its expression in the tissue of NSCLC patients. According to the G0S2 expression levels, patients were divided into high and low expression groups. We further analyzed the clinical characteristics and prognosis in these groups. Results The hypermethylation rate of G0S2 gene in NSCLC patients was high (40%). And the expression of the gene in normal tissues was much higher than those in tumor tissues (P=0.000). There were no significant relationships between the level of expression of this gene and patients' age, sex, smoking history and tumor type cases (P>0.05). Also we couldn't find any relationship between the prognosis and the level of G0S2 gene. Conclusion The expression level of G0S2 gene has some value in the early diagnosis of NSCLC, but there is no obvious value in staging of NSCLC and predicting prognosis. |
