中文摘要:目的 探讨核因子-κB (NF-κB)对A型流感病毒(IAV)性心肌炎小鼠心肌中促炎细胞因子的调控作用。方法 30只8周龄雄性BALB/c小鼠随机分3组:1正常对照组(C组):经鼻吸入15μl生理盐水;2感染对照组(I组):经鼻吸入40空斑形成单位(pfu) IAV;3PDTC组(P组):经鼻吸入40pfu IAV,腹腔注射NF-κB抑制剂吡咯烷二硫代氨基甲酸(PDTC)(10mg/kg),每天1次。感染后第9天处死小鼠,切取心脏组织进行病理及生化检查。结果 IAV感染可诱导心肌中促炎细胞因子白介素(IL)-1β、IL-6及肿瘤坏死因子(TNF)α显著上调(P<0.01,P<0.05),引发心肌急性炎性反应,部分心肌组织坏死。PDTC显著抑制促炎细胞因子在心肌中表达(P<0.01),有效抑制IAV病毒复制(P<0.01),减轻心肌炎性反应。结论 IAV感染心肌组织后可能通过NF-κB信号通路诱导心肌中促炎细胞因子表达上调,抑制NF-κB激活可能为预防和治疗流感病毒性心肌炎提供新的思路。

Regulation of Proinflammatory Cytokine Expression by NF-κB in Viral Myocarditis
Abstract:Objective To investigate the regulatory effects of nuclear factor (NF)-κB on the expression of proinflammatory cytokines in influenza A virus (IAV)-induced myocarditis. Methods Thirty male BALB/c mice of 8 weeks old were randomly divided into 3 groups:1Normal control group (group C):15μL saline with nasal inhalation;2Infection control group (group I):infected intranasally with 40 plaque formation unit (pfu) IAV;3PDTC group (group P):infected intranasally with 40 pfu IAV and injected intraperitoneally with NF-κB inhibitor pyrrolidine dithiocarbamic acid (PDTC)(10mg/kg), once daily. The mice were euthanized at 9 days after infection and the heart tissue was taken for pathological and biochemical examination. Results IAV infection induced significant up-regulation of proinflammatory cytokines (IL) -1β,IL-6 and tumor necrosis factor(TNF) α in myocardium (P<0.01, P<0.05), and triggered acute myocarditis, and some myocardial necrosis. PDTC significantly inhibited the expression of proinflammatory cytokines(P<0.01), and effectively suppressed the IAV replication(P<0.01), and myocardial inflammatory response. Conclusion IAV infection induces up-regulation of proinflammatory cytokines in myocardium might through the NF-κB signaling pathway inhibiting activation of NF-κB may provide new thinking for the prevention and treatment of influenza induced myocarditis.
