中文摘要:目的 观察沉默细胞内氯离子通道蛋白1(chloride channel protein 1,CLIC1)的基因表达后对胃癌BGC-823细胞增值、侵袭、迁移以及周期和凋亡的影响。方法 化学合成针对CLIC1的靶向siRNA (CLIC1 siRNA组),以转染脂质体试剂细胞(MOCK组)和空白细胞(CTRL组)作为对照。用MTT、流式细胞术以及Transwell分别观察细胞的增值、周期、凋亡、侵袭和迁移能力的变化。结果 转染24h、48h和72h后,CLIC1 siRNA组与MOCK组、CTRL组相比,其增殖能力均显著增强(P<0.05);转染48h后CLIC 1siRNA组G2/M期细胞明显增多(P<0.05)、凋亡率显著降低(P<0.05)、侵袭及迁移细胞数明显下降(P<0.05)。结论 CLIC1在胃癌的发生、发展及侵袭迁移中过程发挥重要作用。

Effects of Silencing Chloride Intracellular Channel Ⅰ Gene Expression on the Gastric Cancer Cell
Abstract:Objective To study the effects of silencing CLIC 1 gene expression on proliferation, migraton,invasion, cell cycle and apoptosis of gastric cancer cell line BGC-823. Methods Synthesized siRNA targeting CLIC1 was transfected into BGC-823 cells,while BGC-823 cells transfected with Lipofectamine 2000 and vacant BGC-823 cells were used as controls.Using MTT and flow cytometry,we studied the change of proliferation,poptosis,cell cycle of BGC-823 cells after the expression of clic1 gene was inhabited.At the same time,cell invasion was assessed by transwell chamber. Results Compared CLIC1 siRNA group with MOCK group and CTRL group,after transfecing 24h,48h and 72h with CLIC1 siRNA,the energy of proliferation of BGC-823 cells was significantly increased(P<0.05). After transfecing 48h with CLIC1 siRNA,the number of cells in G2/M period increased (P<0.05), the number of apoptosis cells decreased remarkably(P<0.05). And the number of invasion and migration cells decreased drastically(P<0.05). Conclusion CLIC1 may play an important role in the gastric carcinogenesis and metastasis.
