| 中文摘要:目的 研究BYSL基因在胃癌中的表达情况和临床意义，并探究可能的作用机制。方法 收集胃癌相关生物大数据，对样本中的BYSL基因表达数据及其对应的临床信息进行回顾性分析和生存分析，采用基因集富集分析方法探讨BYSL基因在胃癌中可能的作用机制，采用胃癌及癌旁组织基因芯片数据验证BYSL基因与胃癌发生的相关性。结果 376例胃癌RNA测序样本中，BYSL基因表达与肿瘤分级存在一定相关性（P=0.047），表达越高，肿瘤恶性程度越高，且BYSL基因高表达患者的预后明显差于低表达患者（P=0.000）。基因富集分析提示，BYSL基因高表达的样本中，Myc、E2F1、RPS14、HOXA9等多个肿瘤细胞通路相关基因表达上调。31对胃癌及癌旁组织基因芯片数据也证实，BYSL基因在胃癌中表达明显升高（P=0.000），且具有较大的鉴别诊断价值（AUC=0.958）。结论 BYSL基因表达与胃癌的分级及发展存在一定相关性，具有较大的临床意义。 |

| Clinical Significance of BYSL Expression in Gastric Cancer |

| Abstract:Objective To investigate the clinical significance of BYSL expression and potential mechanism in gastric cancer. Methods Large-scale RNA sequencing data were collected, and retrospective and survival analyses were conducted on the expression profiles of tumor samples and related clinical data. Gene set enrichment analysis was conducted to identify potential mechanism of BYSL in gastric cancer. Finally, the correlation between BYSL expression and gastric cancer was verified by the microarray data of tumor and adjacent tissues. Results In 376 cases of gastric cancer RNA sequencing samples, higher expression of BYSL gene was associated with higher tumor grade (P=0.047). Patients with high BYSL expression showed a poorer prognosis than those with low expression (P=0.000). In gene set enrichment analysis, tumor cellular pathway-related genes (eg. Myc, E2F1, RPS14 and HOXA9) were up-regulated in the samples with BYSL highly expressed. The microarray data of 31 pairs of tumor and adjacent tissues also confirmed a higher expression of BYSL in gastric cancer (P=0.000), with a high diagnostic accuracy (AUC=0.958). Conclusion BYSL expression was related with the grade and prognosis of gastric cancer, and it had a clinical significance. |
