| 中文摘要:目的 探讨不同冻存时间对艾滋病患者血浆外泌体特异性标志物(CD9、CD63、CD81)表达的影响,为优化临床样本保存及标志物选择提供依据。方法 采用Amnis成像流式结合CD9、CD63、CD81荧光抗体标记,对冻存0、6、12个月的艾滋病血浆样本进行定量分析,并通过一级动力学模型评估降解参数。结果 冻存12个月后,CD9、CD63、CD81外泌体占比分别降至初始值的17.3%、21.8%和15.3%,降解符合一级动力学(半衰期分别为5.69、6.49、6.68个月),其中CD81外泌体稳定性最优。冻存12个月后,3种标志物占比相对初始值显著降低(P<0.01)。结论 基于本研究,血浆外泌体标志物的稳定性与其膜蛋白特性密切相关,冻存时间显著影响外泌体标志物的表达。基于降解动力学分析,提出以下保存建议:短期研究(<6个月)可优先选择CD63,兼顾初始丰度与稳定性;长期回顾性分析(≥6个月)推荐以CD81为核心标志物,辅以高敏感度检测技术。 |

| Effect of Cryopreservation Time on Plasma Exosome Markers in AIDS Patients. |

| Abstract:Objective To explore the effects of different cryopreservation times on the expression of exosome-specific markers (CD9, CD63, CD81) in plasma of AIDS patients, and to provide a basis for the optimisation of clinical sample preservation and marker selection. Methods Amnis imaging flow combined with fluorescent antibody labelling of CD9, CD63 and CD81 was used to quantitatively analyse AIDS plasma samples frozen for 0,6 and 12months, and degradation parameters were assessed by a first-order kinetic model.Results After 12months of cryopreservation, the percentage of CD9, CD63 and CD81 exosomes decreased to 17.3%, 21.8% and 15.3% of the initial values, respectively, and the degradation was in accordance with the first-order kinetics (half-lives were 5.69,6.49 and 6.68months, respectively), with CD81 exosomes being the most stable. The percentage of the three markers was significantly reduced after 12months of cryopreservation (P<0.01). Conclusion Based on this study, the stability of plasma exosomal markers is closely related to the characteristics of their membrane proteins, and the cryopreservation time significantly affects the expression of exosome markers. Based on the degradation kinetics analysis, the following preservation recommendations are proposed:CD63 can be preferred for short-term studies (<6months), taking into account the initial abundance and stability; CD81 is recommended as the core marker for long-term retrospective analyses (≥6months), supplemented by high sensitivity detection techniques. |
