| 中文摘要:目的 探讨牛抗菌肽BMAP-28m的生物学功能及其对前列腺癌细胞PC3的增殖和迁移能力的影响。方法 利用生物信息学技术对BMAP-28m进行生物活性的相关分析,并通过MTT实验、光镜下观察细胞形态、划痕实验和克隆形成实验来验证BMAP-28m对前列腺癌细胞PC3的增殖和迁移能力的影响。结果 由27个氨基酸残基组成的阳离子型抗菌肽BMAP-28m的二级结构主要呈α-螺旋结构,同时存在β-结构和无规则卷曲,结构较为稳定。此外,BMAP-28m总体上呈亲水性,可被11种酶切割,存在于细胞内部,具有2个磷酸化位点,且随BMAP-28m的浓度增加,前列腺癌细胞PC3的增殖和迁移能力明显减弱。结论 牛抗菌肽BMAP-28m抑制了前列腺癌细胞PC3的增殖和迁移能力,有望在前列腺癌治疗方面起到积极作用。 |

| Bioinformatics Analysis of Bovine Antimicrobial Peptide BMAP-28m and Its Effect on the Activity of Prostate Cancer Cell PC3. |

| Abstract:Objective To explore the biological function of bovine antimicrobial peptide BMAP-28m and its effect on the proliferation and migration abilities of prostate cancer cell PC3. Methods Bioinformatics techniques were used to conduct relevant analyses on the biological activity of BMAP-28m. The effects of BMAP-28m on the proliferation and migration abilities of prostate cancer cell PC3 were verified through MTT assay, observe the cell morphology under light microscope, scratch assay and colony formation assay. Results The secondary structure of cationic antimicrobial peptide BMAP-28m composed of 27 amino acids, was mainly in the form of α-helix structure, with the presence of β-structure and random coil, and the structure was relatively stable. Additionally, BMAP-28m was generally hydrophilic, can be cleaved by 11 kinds of enzymes, existed inside cells, and has two phosphorylation sites. Moreover, with the increase of BMAP-28m concentration, the proliferation and migration abilities of prostate cancer cell PC3 was significantly weakened. ConclusionThe bovine antimicrobial peptide BMAP-28m inhibits the proliferation and migration abilities of prostate cancer cell PC3, and is expected to play a positive role in the treatment of prostate cancer. |
