中文摘要:目的 探讨区域淋巴结在骨髓间充质干细胞(bone marrow stem cell, BMSC)促进结肠癌细胞增殖中的作用。方法提取和鉴定BMSC,通过细胞计数试剂盒-8(cell counting kit-8,CCK-8)、5-乙炔基-2′-脱氧尿苷(5-ethynyl-2′deoxyuridine,EdU)、平板克隆形成实验研究BMSC对结肠癌细胞(CT26.WT)增殖的影响。选取40只BALB/c小鼠,随机将其分为4组,即不切除区域淋巴结+CT26.WT细胞(A组)、切除区域淋巴结+CT26.WT细胞(B组)、不切除区域淋巴结+BMSC+CT26.WT细胞(C组)、切除区域淋巴结+BMSC+CT26.WT细胞(D组),每组各10只。对各组移植瘤称重并测量体积,通过免疫组织荧光法检测A、B组瘤体内间充质干细胞(mesenchymal stem cell,MSC)分布情况;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法检测小鼠血清中癌胚抗原(carcinoembryonic antigen,CEA)和血管内皮生长因子-C(vascular endothelial growth factor-C,VEGF-C)浓度,免疫组织荧光及苏木精-伊红染色检测瘤体中ki-67的表达。结果 BMSC可以促进结肠癌细胞的增殖(P<0.05);区域淋巴结切除后,瘤体内MSC较未切除组增多;区域淋巴结切除后瘤体的体积、重量以及血清中CEA、VEGF-C、ki-67含量较未切除组增加,并且差异在加入BMSC后更为显著(P<0.05)。结论 BMSC可促进结肠癌细胞的增殖,并且在区域淋巴结切除后增殖作用更为显著,这表明区域淋巴结对MSC具有阻碍作用。

Role of Regional Lymph Nodes in BMSC Promoting the Proliferation of Colon Cancer Cells.
Abstract:Objective To explore the role of regional lymph nodes in in promoting the proliferation of colon cancer cells by bone marrow stem cell (BMSC). Methods BMSC were extracted and identified, and the effect of BMSC on the proliferation of colon cancer cells (CT26.WT) was investigated by cell counting kit-8 (CCK-8), 5-ethynyl-2′-deoxyuridine (EdU), and plate clone formation assay. Forty BALB/c mice were selected and randomly divided into four groups:non-excised regional lymph nodes+CT26.WT cells (group A), excised regional lymph nodes+CT26.WT cells (group B), non-excised regional lymph nodes+BMSC+CT26.WT cells (group C) and excised regional lymph nodes+BMSC+CT26.WT cells (group D), with 10mice in each group. The transplanted tumors of each group were weighed and their volumes were measured, and the distribution of mesenchymal stem cell (MSC) in the tumors of group A and B was detected by immunohistofluorescence; the levels of carcinoembryonic antigen (CEA) and vascular endothelial growth factor-C (VEGF-C) in the serum of mice were detected by enzyme-linked immunosorbent assay (ELISA). The expression of ki-67 in the tumors were detected by immunohistofluorescence and hematoxylin-eosin staining. Results BMSC can promote the proliferation of colon cancer cells (P<0.05); after regional lymph nodes resection, the number of MSC in the tumor was increased compared with the non-resection group; after regional lymph nodes resection, the volume and weight of the tumor, as well as the contents of CEA, VEGF-C and ki-67 in the serum, were increased compared with the non-resection group, and the differences were more significant after the addition of BMSC (P<0.05). Conclusion BMSC can promote the proliferation of colon cancer cells, and the proliferative effect is more significant after regional lymph nodes are resection, suggesting that regional lymph nodes inhibit the MSC.
