| 中文摘要:目的 探究脂肪酸去饱和酶2(fatty acid desaturase 2, FADS2)对胶质母细胞瘤(glioblastoma, GBM)细胞增殖、侵袭和迁移的影响及其作用机制。方法 通过生物信息学分析FADS2在GBM中的表达情况以及对患者预后的影响;通过短发夹RNA(short hairpin RNA, shRNA)干扰U87和U251细胞中FADS2的表达,利用CCK-8法、EdU法和克隆形成实验探究FADS2对GBM细胞增殖的影响;通过Transwell实验分析FADS2对GBM细胞迁移和侵袭的影响,并利用Western blot法分析细胞迁移、侵袭和磷酸肌醇3-激酶(phosphoinositol 3-kinase, PI3K)/蛋白激酶B(protein kinase B, Akt)信号通路相关蛋白的变化情况。结果 FADS2在GBM中的表达水平显著高于肿瘤临近脑组织,且高表达的FADS2与GBM患者的不良预后呈正相关;干扰FADS2的表达抑制了U87和U251细胞的增殖能力,降低了迁移和侵袭能力;Western blot法检测发现,干扰FADS2的表达抑制了PI3K/Akt信号通路,下调N-cadherin和Vimentin蛋白的表达,上调E-cadherin蛋白的表达。结论 FADS2在GBM中高表达,与患者的不良预后呈正相关。干扰FADS2的表达抑制了U87和U251细胞增殖、侵袭和迁移,其机制与抑制PI3K/Akt信号通路相关。 |

| Effects of FADS2 on Proliferation, Invasion and Migration in Glioblastoma Cells and Its Mechanism. |

| Abstract:Objective To investigate the effects of fatty acid desaturase 2 (FADS2) on the proliferation, invasion and migration of glioblastoma (GBM) cells and its mechanism. Methods Bioinformatics analysis was performed to assess FADS2 expression in GBM and its correlation with patient prognosis. The expression of FADS2 in U87 and U251 cells was interfered with short hairpin RNA (shRNA). The effects of FADS2 on GBM cell proliferation were evaluated using CCK-8, EdU, and colony formation assays. Transwell assays were used to analyze the effects of FADS2 on GBM cell migration and invasion. Western blot method was employed to analyze the changes of cell migration, invasion, and the proteins associated with phosphoinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway. Results The expression level of FADS2 in GBM was significantly higher than that in the tumor adjacent to brain parenchyma, and the high expression of FADS2 was positively correlated with the poor prognosis of GBM patients. Interfering with the expression of FADS2 inhibited the proliferation ability of U87 and U251 cells, and reduced their migratory and invasive abilities. Western blot analysis revealed that interfering with the expression of FADS2 inhibited the PI3K/Akt signaling pathway, downregulated the expression levels of N-cadherin and Vimentin proteins, and upregulated the expression levels of E-cadherin protein. Conclusion FADS2 is high expressed in GBM and correlates with poor patient prognosis. Interference with the expression of FADS2 inhibits the proliferation, migration, and invasion of U87 and U251 cells, and the mechanism is associated with the inhibition of the PI3K/Akt signaling pathway. |
