| 中文摘要:目的 通过孟德尔随机化(Mendelian randomization, MR)研究方法分析肠道菌群与哮喘的因果关系。方法 利用公开的全基因组关联研究(Genome-Wide Association Study, GWAS)数据库获取暴露因素肠道菌群及结局哮喘数据集,采用逆方差加权(inverse-variance weighed, IVW)、MR-Egger、加权中位数(weighted median, WM)、简单众数(simple mode, SM)、加权众数(weighted mode, WME)等方法进行MR分析。以OR值评估肠道菌群与哮喘的因果关系,并进行Cochran′s Q检验、MR-Egger截距检验等敏感度分析评判结果的稳健性。结果 IVW法分析结果显示,韦荣球菌属、挑剔真杆菌属、巴尼菌属与哮喘发病风险呈正相关,瘤胃球菌2属、嗜血杆菌属、克里斯滕森菌R7属与哮喘发病呈负相关。敏感度分析均提示结果稳定,不存在异质性和多效性。结论 肠道菌群与哮喘的发生及改善可能存在因果关系,通过调节肠道菌群可能有助于哮喘的防治及改善。 |

| Mendelian Randomization Study on the Causal Relationship between Gut Microbiome and Asthma. |

| Abstract:Objective To analyze the causal relationship between gut microbiota and asthma using Mendelian randomization (MR) research methods. Methods Publicly available Genome-Wide Association Study (GWAS) database were used to obtain datasets of exposure factors with gut microbiota and outcome with asthma. MR analysis was conducted using methods such as inverse-variance weighted (IVW), MR-Egger, weighted median (WM), simple median (SM), and weighted mode (WME). The OR value was used to evaluate the causal relationship between gut microbiota and asthma, and sensitivity analyses such as Cochran′s Q test and MR-Egger intercept test were performed to assess the robustness of the results. Results The results of IVW analysis showed that veillonella group, eubacterium eligens group, and barnesiella group were positively correlated with the risk of asthma, while ruminococcus 2group, haemophilus group, and christensenella R7group were negatively correlated with asthma. Sensitivity analysis indicated that the results were stable, with no heterogeneity or pleiotropy. Conclusion There may be a causal relationship between the gut microbiota and the occurrence and improvement of asthma. Regulating gut microbiota may be helpful for the prevention, treatment and improvement of asthma. |
