中文摘要:目的 探索YKT6在肝细胞癌(hepatocellular carcinoma,HCC)中的表达模式、预后价值、免疫效应和生物学功能。方法 从癌症基因组图谱和基因表达综合数据库中提取有关YKT6表达的原始数据。利用多个数据库分析YKT6在HCC中的表达模式、预后价值,并通过基因本体论(gene ontology,GO)和京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)富集分析探讨YKT6在HCC中的可能的生物学功能,并研究YKT6表达与HCC的免疫浸润及药物敏感度的关系;此外,通过实时荧光定量聚合酶链反应、Western blot法和免疫组化法分析YKT6在HCC细胞和组织中的表达水平。结果 YKT6在HCC细胞和组织中显著上调(P<0.001),这与患者临床病理特征恶化和预后较差相关(P<0.05)。YKT6表达与HCC中的免疫细胞浸润密切相关。GO和KEGG富集分析结果显示,YKT6参与多种肿瘤相关信号通路,主要参与染色体分离、染色体重组、微管蛋白结合和细胞周期等过程,且与多种治疗药物相关。结论 YKT6过表达与HCC进展和不良预后相关,YKT6的表达水平与肿瘤细胞免疫浸润相关,靶向YKT6有望为HCC治疗提供新的思路。

Bioinformatics Analysis of the Expression and Function of YKT6 in Hepatocellular Carcinoma.
Abstract:Objective To explore the expression pattern, prognostic value, immune effects and biological functions of YKT6 in hepatocellular carcinoma (HCC). Methods Raw data on YKT6 expression were extracted from the cancer genome atlas (TCGA) and gene expression omnibus (GEO) databases. The expression pattern and prognostic value of YKT6 in HCC were analyzed using multiple databases, and the possible biological functions of YKT6 in HCC were explored by gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis; and investigated the relationship between YKT6 expression and immune infiltration of HCC and drug sensitivity; moreover, the expression levels of YKT6 in HCC cells and tissues was analyzed by real-time fluorescence quantitative polymerase chain reaction (RT-qPCR), Western blot and immunohistochemistry. Results YKT6 was significantly up-regulated in HCC tissues and cells (P<0.001), which correlated with worsened clinicopathological features and poorer prognosis of patients (P<0.05). The expression of YKT6 was closely associated with immune cell infiltration in HCC. The results of GO and KEGG enrichment analysis demonstrated that YKT6 was involved in a variety of tumor-associated signaling pathways, including those involved in processes such as chromosome segregation, chromosome recombination, microtubule protein binding and cell cycle. YKT6 is also associated with various therapeutic drugs. Conclusion YKT6 overexpression is associated with HCC progression and poor prognosis, and the expression level of YKT6 correlates with the immune infiltration of tumor cells, targeting YKT6 is expected to provide a new concept for the treatment of HCC.
