宫颈癌组织中KDM1A、STIP1、FOXO3A 表达与预后的关系

中文摘要:目的 探讨宫颈癌(cervical cancer,CC)组织中组蛋白赖氨酸特异性去甲基化酶1A[lysine(K)-specific demethylase,KDM1A]、应激诱导磷蛋白1(stress-induced phosphoprotein 1,STIP1)、叉头框蛋白O3A(forkhead box O3A,FOXO3A)表达及其与临床病理特征及预后的关系。方法 招募2020年1月~2022年2月于洛阳市妇幼保健院行手术治疗的CC患者147例。采用免疫组化法检测组织KDM1A、STIP1、FOXO3A蛋白表达,采用Kaplan-Meier生存曲线分析生存情况,采用COX回归分析预后影响因素。结果 CC组织中KDM1A、STIP1蛋白阳性表达率显著高于癌旁正常组织,FOXO3A蛋白阳性表达率显著低于癌旁正常组织(P<0.05)。三者异常表达与国际妇产科联盟(federation international of gynecology and obstetrics,FIGO)分期、淋巴结转移有关(P<0.05)。预后不良组KDM1A、STIP1蛋白阳性表达显著高于预后良好组,FOXO3A蛋白阳性表达显著低于预后良好组(P<0.05)。KDM1A、STIP1阳性表达者3年无进展生存率(59.79%、54.65%)低于阴性表达者(76.00%、80.33%)(Log-Rank χ2分别为5.664、12.870,P<0.05);FOXO3A阳性表达者3年无进展生存率(89.09%)高于FOXO3A阴性表达者(51.09%)(Log-Rank χ2=23.150,P<0.001)。FIGO分期、淋巴结转移及癌组织KDM1A、STIP1、FOXO3A蛋白表达是CC患者预后的影响因素(P<0.05)。结论 KDM1A、STIP1蛋白阳性表达及FOXO3A蛋白阴性表达可能与CC进展相关,是潜在的预后标志物。

Relationship between the Expressions of KDM1A, STIP1, and FOXO3A in Cervical Cancer Tissues and Prognosis.
Abstract:Objective To discuss the expressions of histone lysine (K)-specific demethylase 1A (KDM1A), stress-induced phosphoprotein 1 (STIP1), and forkhead box O3A (FOXO3A) in cervical cancer (CC) tissues, and their relationship with clinical pathological features and prognosis. Methods From January 2020 to February 2022,147 CC patients who underwent surgical treatment in Luoyang Maternal and Child Health Hospital were recruited. Immunohistochemistry was used to detect the expressions of KDM1A, STIP1, and FOXO3A proteins in tissues. Survival was analyzed using Kaplan-Meier survival curves. COX regression analysis was used to analyze prognostic factors. Results The positive rates of KDM1A and STIP1 proteins in CC tissues were significantly higher than those in adjacent normal tissues, while the positive rate of FOXO3A protein was significantly lower (P<0.05). The aberrant expressions of the three proteins were associated with federation international of gynecology and obstetrics (FIGO) staging and lymph node metastasis (P<0.05). The positive expressions of KDM1A and STIP1 proteins in the poor prognosis group were significantly higher than those in the good prognosis group, and the positive expression of FOXO3A protein was significantly lower (P<0.05). The 3-year progression-free survival rate of patients with positive KDM1A and STIP1 (59.79%, 54.65%) was lower than that of patients with negative expression (76.00%, 80.33%) (Log-Rank χ2 were 5.664,12.870, P<0.05); the 3-year progression free survival rate of patients with positive FOXO3A (89.09%) was higher than that of patients with negative FOXO3A (51.09%) (Log-Rank χ2=23.150, P<0.001). FIGO staging, lymph node metastasis, and the expressions of KDM1A, STIP1 and FOXO3A proteins in cancer tissues were prognostic factors for CC patients (P<0.05). Conclusion Positive expressions of KDM1A, STIP1 and negative expression of FOXO3A may be associated with CC progression and serve as potential prognostic markers.
