中文摘要:目的 本研究探讨血红素加氧酶-1与氟尿嘧啶诱导食管癌细胞凋亡的关系。 方法 培养ECa109食管鳞癌细胞,实验组分别加入正常培养基,含20μmol/L、80μmol/L锌原卟啉(ZnppIX)的培养基培养12h,后更换为含100μg/ml 5-FU组培养;对照组予正常培养基培养;共培养96h后检测各组细胞凋亡情况。 结果 在凋亡早期细胞中(Annexin V+/PI-),20μmol/L ZnppIX组(25.63%±6.52%)及80μmol/L ZnppIX(22.48%±5.59%)组显著高于5-FU组(12.89%±4.42%,P均<0.05)。在中晚期凋亡细胞(Annexin V+/PI+)中,20μmol/L ZnppIX组(25.17%±5.53%)显著低于5-FU组(39.89%±6.66%)及80μmol/L ZnppIX组(41.95%±2.92%,P均<0.01)。80μmol/L ZnppIX组caspase-3的活化率显著高于正常细胞(P<0.05)。 结论 ZnppIX抑制HO-1表达后,可以显著增加5-FU诱导的Eca109食管癌细胞凋亡率,caspase-3参与了其诱导细胞凋亡的过程。

Influence of Heme Oxygenase-1 on Apoptosis Induced by Fluorouracil in ECa109 Cells
Abstract:Objective To exploe relationchip teetmeen the expression of HO-1 and flwrouravil in esophageal carcinoma Methods ECa109 cells were trained 12 hours by nomal medium, 20μmol/L and 80μmol/L concentrations of ZnppIX medium, then trained by 100μg/ml 5-FU to 96 hours totally. The apoptosis was detected in all experimental groups marked by Annexin Ⅴ, PI and caspase-3. Results In early apoptosis, 20μmol/L ZnppIX group (25.63%±6.52%) and 80μmol/L ZnppIX (22.48%±5.59%) group was significantly higher than the 5-FU group (12.89%±4.42%) (P<0.05). In late apoptotic, 20μmol/L ZnppIX group (25.17%±5.53%) was significantly lower than the 5-FU group (39.89%±6.66%) and 80μmol/L ZnppIX group (41.95%±2.92%) (P all <0.01). The caspase-3 activation rate in 80μmol/L ZnppIX group was significantly higher than normal cells(P<0.05). Conclusion ZnppIX significantly increased the apoptosis of ECa109 cells caused by 5-FU. Caspase-3 took a part in this apoptosis.
