中文摘要:目的 探讨烟雾主要成分苯并芘(BaP)对RAW264.7单核-吞噬细胞中基质金属蛋白酶(MMPs)的影响,为研究吸烟与腹主动脉瘤的关联提供相关依据。方法 应用免疫印迹法观察BaP对体外培养的RAW264.7单核-吞噬细胞基质金属蛋白酶-2/9/12(MMP-2/9/12)的影响,并验证其是否通过NF-κB信号通路。结果 BaP暴露使培养巨噬细胞NF-κB活化,同时伴随MMP-9、MMP-12表达升高,而PDTC,核转录因子NF-κB的抑制剂,可抑制NF-κB活化,同时伴随MMP-9、MMP-12表达下降。结论 BaP可通过NF-κB通路诱导RAW264.7单核-吞噬细胞MMP-9、MMP-12表达升高。 |

Abstract:Objective To explore the effect of benzopyrene(BaP),the maincomponents of smoke,on matrix metalloproteinase(MMPs) of RAW246.7 mononuclear macrophage. Methods We observed the influence of BaP on matrix metalloproteinase-2/9/12(MMP-2/9/12) of RAW246.7 mononuclear macrophage in vitro by western blotting and verified if it happened through the NF-κB signaling pathway. Results BaP caused NF-κB activation and expression of MMP-12, MMP-9 increased in cultured macrophages.While PDTC,an inhibitor of nuclear transcription factor NF-κB,caused the inhibition of NF-κB activation and expression of MMP-12 and MMP-9 decreased. Conclusion BaP can induce the expression of MMP-9 and MMP-12 in RAW246.7 cell by NF-κB pathway. |
