芬戈莫德对血管紧张素Ⅱ灌注大鼠肾脏炎症的影响

中文摘要:目的 探讨芬戈莫德(Fingolimod,FTY720)对血管紧张素Ⅱ(angiotensin Ⅱ,AngⅡ)灌注大鼠肾脏炎症改变的影响。方法 36只雄性SD大鼠随机分为3组:①模型组12只,即AngⅡ输注组,皮下置入含AngⅡ的渗透性微量泵,持续灌注14天或28天[AngⅡ:400ng/(kg·min)];②干预组12只,即AngⅡ+FTY720干预组,在模型组的基础上,FTY720以0.5g/(kg·d)灌胃14天或28天;③对照组12只,以等量的生理盐水灌胃14天或28天。每周称量体重,留24h尿液,分别于第14天、28天分批处死各组大鼠,经心脏采血,并留取肾脏标本。放射免疫法检测血清AngⅡ及肾匀浆组织中AngⅡ的浓度。尿蛋白与血生化分析。免疫组织化学法检测炎性因子TNF-α、IL-6在肾脏中的表达。结果(1)AngⅡ可导致血肌酐、尿素氮水平升高,尿蛋白明显增加,炎性因子TNF-α、IL-6在肾脏组织中的表达水平升高,差异有统计学意义;(2)干预组的肾脏病理损伤较模型组减轻,血肌酐、尿素氮水平明显降低,尿蛋白明显减少,肾脏组织中炎性因子TNF-α、IL-6水平明显降低。结论 FTY720可减轻AngⅡ诱导的大鼠肾脏的病理损伤,可能是通过减少炎性因子的表达水平发挥了减轻尿蛋白,改善肾功能的作用。

Effect of Fingolimod on Renal Inflammation in Angiotensin Ⅱ-infused Rat Model
Abstract:Objective To investigate the effect of Fingolimod(FTY720) on renal inflammation in angiotensin Ⅱ-infused rat model. Methods Thirty-six male SD rats were randomly divided into 3 groups:① AngⅡ-infused group:AngⅡ osmotic micro-pump implanted subcutaneously for 14 days or 28 days[AngⅡ:400ng/(kg·min)];② Intervented group(AngⅡ-infused group+FTY720):Based on the AngⅡ-infused group, treated with FTY720 as a concentration of 0.5g/(kg·d) for 14 days or 28 days;③ Control group:with the same amount of normal saline for 14 days or 28 days. 24h urine, rat serum and kidney tissue were collected on day 14,28 respectively before the rats were sacrificed.The concentration of AngⅡ in serum and renal homogenate were detected by radioimmunoassay. Urine protein was measured by salicylic acid method. The expression levels of TNF-α and IL-6 in kidney tissue were analyzed by immunohistochemistry. Results AngⅡ could increase the levels of serum creatinine and urea nitrogen. Meanwhile, AngⅡ could increase the levels of inflammatory factors TNF-α and IL-6 in kidney tissue. Compared with the model group, the renal pathological injury was significantly alleviated in the intervented group. Serum creatinine and urea nitrogen levels in the intervention group were significantly lower than those in model group. The levels of inflammatory factors TNF-α and IL-6 in the intervention group were also significantly decreased compared to model group. Conclusion FTY720 can reduce the pathological damage of the kidney in rats induced by AngⅡ, which may decrease the urinary protein and improve the renal function by reducing the expression levels of inflammatory factors.
