高同型半胱氨酸血症兔致动脉粥样硬化的实验研究

中文摘要:目的 复制可靠的兔高同型半胱氨酸血症(HHcy)致动脉粥样硬化(AS)模型。方法 采用L-蛋氨酸皮下注射法建立HHcy家兔模型,并设生理盐水组为对照,比较两组注射前、注射后4、8、12周血清中Hcy、TC、TG、HDL-C、LDL-C值,实验结束后超声观察腹主动脉狭窄程度及病理切片观察AS形成的情况。结果 皮下注射L-蛋氨酸4周后血中Hcy明显高于正常组(P<0.05),HHcy模型形成;造模8、12周TC、TG水平显著升高(P<0.05);造模12周模型组LDL-C水平显著升高(P<0.05);造模12周后腹主动脉超声与病理均提示有明显的AS征象。结论 L-蛋氨酸皮下注射80mg/(kg·d)4周可建立HHcy模型,12周时动脉粥样硬化表现明显。

Experimental Study on High Homocysteine Induced Rabbit Atherosclerotic Lesions
Abstract:Objective To replicate animal model of high homocysteine induced rabbit atherosclerotic lesions.Methods Rabbits were randomly divided into 2 groups:the model group and the control group. The model groups were injected L-methionine respectively, while the control group was given normal saline. The levels of Hcy in serum were analyzed before injection, 4 weeks, 8 weeks and 12 weeks after injection in each group. The formation of artherosclerosis was observed through type-B ultrasonic and pathological section. Moreover, the levels of Hcy, TC, TG, HDL-C and LDL-C in serum of each group were determined.Results After 4 weeks of injection, the levels of Hcy in serum in model groups were significantly higher than the control group (P<0.05). After 8 weeks and 12 weeks, the levels of TC and TG were significantly increased (P<0.05). After 12 weeks, the levels of LDL-C were significantly increased (P<0.05). After 12 weeks, the abdominal aorta has obvious signs of atherosclerosis with type-B ultrasonic and pathological section.Conclusion When administrated with L-methionine injection[80mg/(kg·d)], rabbit can be made into the homocystinemia model in the 4th weeks and artherosclerosis can be developed on the 12th week
